PPI claims blamed for increase in bank complaints
New figures released through the Financial Services Authority (FSA) show that the number of complaints received by financial institutions has risen by three percent during the first six months of this year with PPI being seen as responsible.The FSA figures reveal that the total number of cases which related to payment protection insurance was up by 23% to over half a million and banks are braced for this figure to increase further as more people realise that they have been mis-sold the insurance, which had been sold to customers so as to cover repayments in case the borrower becomes unemployed or falls ill. Banks already have put aside over £7bn to pay compensation for those customers’ who had been mis-sold policies, yet there is now evidence that banks are coming to terms with the number of complaints coming their way and they are now taking steps to attempt to address the problem earlier on in the process. Some banks have previously acknowledged that they have to make improvements in the manner they handle complaints and those, including Lloyds and Santander, which attract the second and third highest amount of complaints, already have begun to put new systems into position. Barclays, again attracted most complaints receiving 251,563, though it was quick to say that this was over 14% down on the same period in 2010. A spokesman added that the number of complaints was due to PPI that is affecting the whole sector and that the banking group had generally made good progress in reducing the number, achieving a decrease of almost a third in banking complaints.The rise in the total number of people taking further action is mainly due to the decision, in May, from the banking industry to drop its legal challenge on whether or not the new FSA rules on PPI mis-selling claims were able to be applied retrospectively. This has led to an expected total bill of £9bn with over three million people expecting to receive compensation.A ruling from the FSA last year determined that financial companies were required to publish the total number of complaints they received every six months and also stated that complaints were to be defined as oral or written expressions of dissatisfaction and had to be recorded whether or not they were considered to be justified.The chief executive of Which? has said that the FSA figures show that the market isn’t working satisfactorily and that it ought to be made simpler for dissatisfied customers to change banks. Peter Vicary-Smith said that high street banks had a poor track record for customer satisfaction and that the right way to produce a more appealing market for customers would be for the regulator to encourage competition by helping the switching process from bank to bank. He said this would then provide financial institutions with a simple choice, either look after their clients or face the risk of losing them altogether.The FSA figures were part of a wider survey on banking complaints which revealed that, while Barclays received the most complaints, Lloyds received the most of any banking group with 317,332 complaints across its group, which includes Lloyds TSB, Halifax, Bank of Scotland and Cheltenham & Gloucester. One bit of good news contained in the figures was that all major brands were dealing with complaints within the eight weeks stipulated by the FSA. Paul Clark, chief executive of Charter UK, which develops complaint software, claimed that banks were improving in this area and that increasingly they were starting to realise how their name and reputation can be damaged. He added that with better systems in place, together with a more proactive approach, the businesses concerned may start to see the positives in using the feedback which comes out of complaints.

Computational and experimental study of the acoustic startle response and prepulse inhibition. $49.99 We analyzed different computational models of the acoustic startle response (ASR) and prepulse inhibition (PPI) in order to better understand and characterize the underlying neural mechanisms of these phenomena, and to explain the role of specific brain areas in different experimental manipulations. The models include a neural network developed from behavioral results, and a realistic model based on neurobiological data. The neural network model incorporates excitatory and facilitatory pathways activated by the positive value of changes in noise level in the environment, and an inhibitory pathway activated by the absolute value of changes in noise level. Whereas excitation and facilitation are exponential functions, inhibition is a linear function of the input noise. The model describes many properties of the ASR and PPI, the effects of specific brain lesions, and correctly predicts how PPI depends on pulse intensity. In addition, we experimentally establish the magnitude of startle and PPI as a function of pulse, prepulse, and background intensity, and test the model predictions regarding an inverted-U function that relates startle to the intensity of the background noise. The realistic model includes an excitatory pathway that represents neurons projecting directly from the cochlear nucleus (CN) to the caudal pontine reticular nucleus (PNC), and an inhibitory pathway that replicates the CN-inferior colliculus (IC)-pedunculopontine tegmental nucleus (PPT)-PNC neural connections. Even though this model is able to capture some behavioral properties of the ASR and PPI, the simulated results produced with the neural network reproduce better the experimental data. However, the realistic model can provide physiologically plausible explanations for behavioral results, such as prepulse facilitation (PPF). The role of dopaminergic mechanisms in the ASR and PPI attenuation in rats caused by the introduction of novel changes in environment illumination is investigated. |

Computational and experimental study of the acoustic startle response and prepulse inhibition. $49.99 We analyzed different computational models of the acoustic startle response (ASR) and prepulse inhibition (PPI) in order to better understand and characterize the underlying neural mechanisms of these phenomena, and to explain the role of specific brain areas in different experimental manipulations. The models include a neural network developed from behavioral results, and a realistic model based on neurobiological data. The neural network model incorporates excitatory and facilitatory pathways activated by the positive value of changes in noise level in the environment, and an inhibitory pathway activated by the absolute value of changes in noise level. Whereas excitation and facilitation are exponential functions, inhibition is a linear function of the input noise. The model describes many properties of the ASR and PPI, the effects of specific brain lesions, and correctly predicts how PPI depends on pulse intensity. In addition, we experimentally establish the magnitude of startle and PPI as a function of pulse, prepulse, and background intensity, and test the model predictions regarding an inverted-U function that relates startle to the intensity of the background noise. The realistic model includes an excitatory pathway that represents neurons projecting directly from the cochlear nucleus (CN) to the caudal pontine reticular nucleus (PNC), and an inhibitory pathway that replicates the CN-inferior colliculus (IC)-pedunculopontine tegmental nucleus (PPT)-PNC neural connections. Even though this model is able to capture some behavioral properties of the ASR and PPI, the simulated results produced with the neural network reproduce better the experimental data. However, the realistic model can provide physiologically plausible explanations for behavioral results, such as prepulse facilitation (PPF). The role of dopaminergic mechanisms in the ASR and PPI attenuation in rats caused by the introduction of novel changes in environment illumination is investigated. |

Nanotechnology in developing multifunctional non-viral gene delivery systems. $49.99 Small interfering RNA (siRNA) has emerged as a powerful technology for sequence specific suppression of genes in mammalian cells and has broad applications ranging from functional gene analysis to targeted therapy. However, its clinical success is still limited because of the inherent instability and poor intracellular internalization of siRNA. Hence, the aim of the thesis study is to develop a safe non-viral siRNA delivery vehicle for effective cancer therapy. To this end, we took advantage of various nanomaterials and nanotechnologies, combined with the traditional biological techniques to reach our goal. Chapter 1 gives a background of this research and Chapter 2 summarizes the experimental procedures and techniques. Chapter 3 evaluates different generations of PPI dendrimers as potential siRNA delivery agents for cancer therapy. We demonstrated that the higher generations of PPI dendrimers were able to effectively complex siRNA into discrete nanoparticles, which resulted in a sufficient knockdown of targeted gene expression in human cancer cells. The mechanism of siRNA complexation by PPI dendrimers was also investigated in Chapter 4. Results suggest that PPI dendrimers first 'zip' the siRNA molecules by electrostatic interactions to form extended nanofibers, followed by secondary folding into nanoparticles. Understanding this complexation pathway is crucial in the rational design of more effective vectors for therapeutic siRNAs. Besides siRNA complexation, attempts to develop effective non-viral siRNA vectors for in vivo delivery are hampered by difficulties that include extracellular stability, siRNA intracellular availability and target cell specificity. Chapter 5 focuses on the development of a non-viral gene delivery vector to address the above mentioned problems. Understanding cellular function through the analysis of protein interaction networks. $49.99 A major challenge of post-genomic biology is understanding the complex networks of interacting genes, proteins and small molecules that give rise to biological form and function. Advances in whole-genome approaches are now enabling us to characterize these networks systematically, using procedures such as the two-hybrid assay and protein co-immunoprecipitation to screen for protein-protein interactions (PPI). Large protein networks are now available for many species like the baker’s yeast, worm, fruit fly and the malaria parasite P. falciparum. These data also introduce a number of technical challenges: how to separate true protein-protein interactions from false positives; how to annotate interactions with functional roles; and, ultimately, how to organize large-scale interaction data into models of cellular signaling and machinery. Further, as protein interactions form the backbone of cellular function, they can potentially be used in conjunction with other large-scale data types to get more insights into the functioning of the cell. In this dissertation, I try to address some the above questions that arise during the analysis of protein networks.;First, I describe a new method to assign confidence scores to protein interactions derived from large-scale studies. Subsequently, I perform a benchmarking analysis to compare its performance with other existing methods. Next, I extend the network comparison algorithm, NetworkBLAST, to compare protein networks across multiple species. In particular, to elucidate cellular machinery on a global scale, I performed a multiple comparison of the protein-protein interaction networks of C. elegans, D. melanogaster and S. cerevisiae . This comparison integrated protein interaction and sequence information to reveal 71 network regions that were conserved across all three species and many exclusive to the metazoans. I then applied this technique to the analysis of the protein network of the malaria pathogen Plasmodium falciparum |
